ACTIVITY TEST OF GRAPE LEAF EXTRACT OINTMENT AGAINST THE HEALING OF ARTIFICIAL WOUNDS OF RAT SKIN INDUCED DIABETES STREPTOZOTOSIN

Abstract

The plant sembung rambat (Mikania micrantha Kunth.) is one of the weeds in Indonesia. Traditionally, the community has used and has been empirically proven that the sembung rambat plant as a wound medicine, for example wounds in people with diabetes, so it is likely that it can be developed into an alternative wound healing medicine. The purpose of this study was to determine the ointment preparation containing ethanol extract of sembung rambat leaves can heal artificial wounds on the skin of male rats that have been induced diabetes with streptozotocin. This research was conducted with experimental method by using test material of sembung rambat leaves made into simplisia, then extracted by percolation using 80% ethanol solvent and tested for phytochemical screening, making ointment preparations from extracts with various concentrations, tested the physical quality of the preparation including homogeneity, stability, pH, irritation, liking. Then tested for artificial wound healing in diabetic male rats induced with streptozotocin. The results of phytochemical screening showed that sembung rampat leaves, simplisia, and ethanol extract contained alkaloids, flavonoids, tannins, saponins, steroids and glycoside compounds. The ointment preparation of ethanol extract of semung rambat leaves at a concentration of 2.5%, 5%, 7.5%, homogeneous, stable until the 4th week, pH around 6.16, no irritation on the skin of volunteers. Has the effectiveness of artificial wound healing in diabetic rats induced with streptozotocin. On day 19, the percentage of wound healing was not significantly different between the administration of EEDSr 7.5% ointment and gentamicin, day 21 was not significantly different between the administration of EEDSr 5%, EEDSr 7.5%, and gentamicin. So it is concluded that giving 5% EEDSr gives the best results, because on day 21 it is not significantly different from 7.5% and gentamicin.
